124I-F8IL10 is an investigational radiolabeled immunocytokine and biologic fusion protein developed for positron emission tomography (PET) imaging. It consists of the fully human fusion protein F8-IL10 (also known as Dekavil or PF-06687234) labeled with the positron-emitting radioisotope iodine-124 (124I). The F8 antibody fragment of the fusion protein selectively targets the extra-domain A (EDA) of fibronectin, a marker of tissue remodeling and angiogenesis that is highly expressed at sites of active inflammation. The interleukin-10 (IL-10) cytokine component acts as an anti-inflammatory mediator. Originally developed by Philogen and licensed to Pfizer, 124I-F8IL10 has been evaluated in Phase 0/1 clinical trials (conducted at VU University Medical Center and other sites) as a diagnostic imaging tracer to assess pharmacokinetics, biodistribution, and inflammatory-site targeting in patients with active rheumatoid arthritis and inflammatory bowel disease (including Crohn's disease and ulcerative colitis).
