Overview

**124I-omburtamab** is a theranostic radiopharmaceutical consisting of the murine monoclonal antibody omburtamab, which targets the tumor-associated antigen B7-H3 (CD276), conjugated with the radioisotope iodine-124. It is designed for targeted delivery of radioactivity to tumors expressing B7-H3, providing both imaging and therapeutic effects. Most clinical research has focused on pediatric brain tumors, especially diffuse intrinsic pontine glioma (DIPG), employing direct infusion into the brain via convection-enhanced delivery. The agent is under investigation primarily for its ability to maximize radiation at tumor sites while minimizing systemic exposure. The developer is Y-mAbs Therapeutics, with the original antibody developed at Memorial Sloan Kettering Cancer Center. 124I-omburtamab is distinct from 131I-omburtamab, but shares the same antibody backbone.
