Drug intelligence / Profile preview
12A12 is a cleavage-specific monoclonal antibody (mAb) that selectively targets and neutralizes the neurotoxic, N-terminal 20-22 kDa tau fragment(s) (NH2htau) in the brain without cross-reacting with full-length physiological forms of the tau protein. Developed by researchers at the European Brain Research Institute (EBRI) and the National Research Council (CNR) of Italy, 12A12 is designed to intercept the co-pathogenetic link between amyloid-beta (Aβ) and tau. In preclinical models of Alzheimer's disease (such as Tg2576 and 3xTg mice), systemic administration of 12A12 has been shown to reduce both pathological tau and amyloid-beta accumulation, improve cognitive and visuo-spatial memory, restore synaptic plasticity, and mitigate neuroinflammation. It is currently in preclinical development for the treatment of Alzheimer's disease.
