Overview

12G2-vt9 (also known as 12G2-variant 9 or S-V9) is a humanized monoclonal antibody designed to target and neutralize adipocyte fatty acid-binding protein (A-FABP, also known as FABP4). Developed by researchers at the University of Iowa and its spinout BMImmune, 12G2-vt9 is being investigated for the treatment of breast cancer, particularly obesity-associated and triple-negative breast cancer (TNBC). FABP4 is a lipid chaperone that is elevated in circulation during obesity and is known to promote cancer stemness, tumor growth, and metastasis. By blocking extracellular FABP4, 12G2-vt9 disrupts the FABP4/ALDH1 signaling axis, reduces cancer stemness, and impairs tumor cell mitochondrial energy metabolism. In preclinical models, 12G2-vt9 has demonstrated significant efficacy in inhibiting mammary tumor growth and lung metastasis.
