Overview

1312-CAR-T is an experimental second-generation chimeric antigen receptor (CAR) T-cell therapy designed to target the B-cell activating factor receptor (BAFFR), a member of the tumor necrosis factor (TNF) receptor superfamily primarily expressed on B cells. Developed by Guangzhou Bio-gene Technology and DIMA Biotechnology, the therapy incorporates a specific scFv derived from the 1312 antibody clone, linked to 4-1BB and CD3ζ signaling domains. BAFFR is highly expressed in various B-cell malignancies and plays a role in B-cell maturation, making it a target for both hematologic cancers and autoimmune diseases. Preclinical data presented at ASCO 2025 demonstrated that 1312-CAR-T exhibits potent cytotoxicity and cytokine release against Raji tumor cells in vitro and effectively clears tumors in mouse models. It also showed potential in reducing dsDNA-IgG levels in systemic lupus erythematosus (SLE) models, suggesting therapeutic utility in autoimmune conditions.
