Overview

1313-CAR-T is a second-generation chimeric antigen receptor (CAR) T-cell therapy targeting the B-cell Activating Factor Receptor (BAFFR). Developed by Guangzhou Bio-gene Technology and DIMA Biotechnology, it utilizes a specific scFv derived from the 1313 antibody clone, coupled with a 4-1BB costimulatory domain and a CD3ζ signaling domain. BAFFR is almost exclusively expressed on B cells and is highly expressed in B-cell malignancies and involved in autoimmune disease pathogenesis. In preclinical studies, 1313-CAR-T demonstrated effective in vitro cytotoxicity against Raji tumor cells, although it was noted to have lower cytokine release and cytotoxicity compared to the 1312-CAR-T variant. It is being investigated for the treatment of B-cell acute lymphoblastic leukemia (B-ALL), non-Hodgkin lymphoma (NHL), and autoimmune diseases such as systemic lupus erythematosus (SLE).
