131I-ELP is a research-stage radiotherapeutic agent consisting of a thermally responsive elastin-like polypeptide (ELP) biopolymer conjugated with iodine-131 (131I) radionuclides. Designed for localized brachytherapy, the agent is administered as a liquid that undergoes a phase transition at body temperature to form a stable, insoluble depot within the tumor. This depot provides sustained, localized beta-radiation delivery while minimizing systemic exposure. Preclinical studies, primarily conducted at Duke University, have demonstrated significant efficacy in pancreatic cancer models, particularly when used in combination with systemic chemotherapy like nab-paclitaxel. The ELP carrier allows for a high concentration of the radionuclide to remain at the injection site, overcoming the limitations of rapid clearance typically seen with small-molecule radiopharmaceuticals.
