131I-huA33 is a humanized monoclonal antibody (huA33) conjugated with radioactive iodine-131. The antibody targets the A33 antigen, which is highly expressed on more than 95% of colorectal cancers and in normal intestinal epithelium but not in most other tissues. By binding to the A33 antigen on tumor cells and delivering targeted radiation via iodine-131, it enables selective radioimmunotherapy for metastatic colorectal cancer. Clinical studies have demonstrated that 131I-huA33 achieves specific tumor targeting with minimal uptake in non-tumor tissues except transiently in bowel epithelium. It has been evaluated both as monotherapy and in combination with chemotherapy (notably capecitabine), showing promising safety and efficacy profiles at defined maximum tolerated doses[2][3][8].
