131I-IFPABZA (131I-N-(4-((2-(diethylamino)ethyl)carbamoyl)-2-iodo-5-methoxyphenyl)-5-fluoropicolinamide) is a radioiodinated theranostic agent developed for the management of malignant melanoma. It belongs to a class of fluorinated picolinamide-benzamide derivatives designed to exploit the high affinity of benzamides for melanin, a pigment prevalent in melanoma tumors. The incorporation of Iodine-131 allows the compound to serve as both a SPECT imaging probe and a targeted radiotherapeutic, delivering ionizing radiation directly to pigmented cancer cells. Preclinical evaluations in B16F10 melanoma-bearing mice have demonstrated that 131I-IFPABZA exhibits high tumor uptake, prolonged retention, and favorable tumor-to-organ ratios, particularly compared to its nicotinamide counterpart. Its pharmacokinetic profile is characterized by rapid renal clearance, which helps minimize radiation exposure to non-target tissues.
