13F04 + PNU-159682 is an experimental antibody-drug conjugate (ADC) designed to target the L1 cell adhesion molecule (L1CAM), a transmembrane protein that serves as a selective marker for metastasis stem cells (MetSCs). The ADC consists of the monoclonal antibody 13F04, which binds with high affinity and specificity to the extracellular domain of human L1CAM, conjugated to the highly potent anthracycline derivative PNU-159682. PNU-159682 is a metabolite of nemorubicin that acts as a potent DNA intercalator and topoisomerase II inhibitor, inducing cytotoxic cell death in targeted tumor cells. By specifically eliminating L1CAM-expressing MetSCs, which are associated with phenotypic plasticity and resistance to conventional chemotherapy, this ADC aims to prevent metastatic growth and tumor relapse. Preclinical studies in models of triple-negative breast cancer and lung adenocarcinoma have demonstrated robust tumor control and extended survival, supporting its development as a therapeutic agent for advanced solid tumors.
