Overview

1400W is a highly selective, slow, tight-binding, and largely irreversible small molecule inhibitor of inducible nitric oxide synthase (iNOS). It exhibits over 5000-fold selectivity for iNOS compared to endothelial nitric oxide synthase (eNOS), and more than 200-fold selectivity over neuronal nitric oxide synthase (nNOS). The inhibition is time-dependent and either irreversible or extremely slowly reversible. In preclinical models, 1400W has demonstrated efficacy in reducing nitrooxidative stress, neuroinflammation, and neurodegeneration following organophosphate nerve agent exposure as well as in other models of brain injury. Its mechanism involves competitive inhibition with L-arginine at the iNOS active site. Developed originally by Glaxo Wellcome researchers for research purposes, it is widely used as a tool compound to study the role of iNOS in various disease processes including neurodegenerative disorders and inflammation[1][3][5][8].
