141-A is a highly potent and selective allosteric inhibitor of the p38α MAPK-activated protein kinase-2 (MK2) axis, developed by Changchun Genescience Pharmaceuticals (GenSci). Unlike traditional p38α inhibitors that block all downstream signaling, 141-A specifically inhibits the p38α-mediated activation of MK2 while sparing other pathways such as ATF2 and PRAK. This selective mechanism is designed to reduce the production of pro-inflammatory cytokines (including TNFα, IL-1β, IL-6, IL-8, and IL-17) while potentially avoiding the side effects associated with broad p38α inhibition. Preclinical data in models of adjuvant-induced arthritis, psoriasis, and MSU-induced inflammation have demonstrated significant efficacy and a favorable safety profile. As of early 2024, the compound is in IND-enabling studies with a Phase 1 clinical trial planned for early 2025.
