143G03-PNU-159682 ADC is an experimental antibody-drug conjugate (ADC) designed to target the L1 cell adhesion molecule (L1CAM), a transmembrane protein highly expressed on metastasis stem cells (MetSCs) across various solid tumors. The ADC is composed of the monoclonal antibody 143G03, which exhibits high affinity and specificity for the extracellular domain of human L1CAM, conjugated to the extremely potent anthracycline derivative PNU-159682. PNU-159682 is a metabolite of nemorubicin that acts as a powerful DNA intercalator and cytotoxic agent. By specifically targeting L1CAM, the ADC aims to eliminate drug-resistant cell populations responsible for metastatic growth and cancer relapse. Preclinical studies have demonstrated significant efficacy in mouse models of triple-negative breast cancer and lung adenocarcinoma, showing robust tumor control and improved survival rates.
