14A10-PNU-159682 ADC is an experimental antibody-drug conjugate (ADC) designed to target the L1 cell adhesion molecule (L1CAM), a transmembrane protein overexpressed in various metastatic solid tumors and specifically on drug-resistant metastasis stem cells (MetSCs). The ADC consists of the 14A10 monoclonal antibody, which binds with high affinity to the extracellular domain of L1CAM, conjugated to PNU-159682, a highly potent anthracycline derivative and metabolite of nemorubicin. PNU-159682 acts as a powerful DNA-intercalating agent and topoisomerase II inhibitor, inducing potent cytotoxicity. By targeting L1CAM, the ADC delivers this cytotoxic payload directly to tumor cells responsible for metastatic spread and chemoresistance. Preclinical studies have demonstrated robust tumor control and extended survival in mouse models of triple-negative breast cancer and lung adenocarcinoma.
