14B7 is a murine monoclonal antibody of the IgG1 isotype that specifically targets N-glycolylneuraminic acid (NeuGc)-containing gangliosides, most notably NeuGcGM3. This carbohydrate antigen is considered a tumor-specific neoantigen; it is absent in normal human tissues but highly expressed in various malignancies, including non-small cell lung cancer (NSCLC), breast cancer, and melanoma, due to the metabolic incorporation of dietary NeuGc or alterations in the sialic acid pathway in tumor cells. The mechanism of action for 14B7 involves direct binding to NeuGcGM3 on the tumor cell surface, which induces a rapid, non-apoptotic cell death process resembling oncosis, characterized by large membrane pores and loss of cytoplasmic integrity. Additionally, 14B7 can mediate antibody-dependent cellular cytotoxicity (ADCC). The antibody has been investigated in Phase I and II clinical trials as a passive immunotherapy and has also served as the foundational 'Ab1' antibody for the development of the anti-idiotype vaccine racotumomab (Vaxira), which mimics the NeuGcGM3 epitope to induce an active immune response in patients. 14B7 has also been utilized in radiolabeled forms for the diagnostic imaging of NeuGcGM3-positive tumors.
