Overview

**14G2A** is a murine monoclonal antibody of the IgG2a isotype that specifically targets ganglioside GD2, a sialic acid-containing glycosphingolipid highly expressed on certain tumors of neuroectodermal origin, notably neuroblastoma and melanoma[2][3][6]. The antibody was derived as an isotype switch variant from the parental IgG3-producing hybridoma 14.18 and possesses identical antigen-binding reactivity[2][6].\nIts mechanism of action includes direct binding to the extracellular GD2 epitope on tumor cells, leading to tumor cell death through several immune effector functions: antibody-dependent cell-mediated cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), and, in some contexts, induction of apoptosis[4][5][6]. It has shown efficacy in preclinical and early clinical studies, particularly for neuroblastoma, and is considered the prototype for subsequent clinical anti-GD2 antibodies such as ch14.18 (dinutuximab)[6].\nPrimary indication is for the immunotherapy of neuroblastoma, with additional research targeting melanoma and other GD2-positive tumors[1][3][4][6].
