**14G2a-gelonin** is an experimental GD2-directed immunotoxin consisting of the anti-disialoganglioside GD2 monoclonal antibody 14G2a chemically conjugated through an SPDP linker to gelonin, a plant-derived ribosome-inactivating protein. The antibody targets GD2-expressing neuroectodermal tumor cells, while internalized gelonin inhibits protein synthesis through ribosomal RNA depurination. In published in-vitro work, the conjugate retained 14G2a immunoreactivity, complement-dependent cytotoxicity, and antibody-dependent cellular cytotoxicity, and showed direct cytotoxicity against human melanoma cell lines with substantially greater activity than unconjugated gelonin.
