14g2a.zeta GD2-CAR EBV-CTLs is a first-generation chimeric antigen receptor (CAR) T-cell therapy specifically designed to target the disialoganglioside GD2, which is highly expressed on neuroectodermal tumors. The therapeutic construct consists of Epstein-Barr virus-specific cytotoxic T-lymphocytes (EBV-CTLs) that have been retrovirally transduced with a CAR comprising a single-chain variable fragment (scFv) derived from the 14G2a monoclonal antibody, fused to the CD3-zeta signaling endodomain. By using EBV-specific CTLs as the cellular substrate, the therapy intends to utilize the native T-cell receptor (TCR) to provide endogenous costimulation through interactions with EBV-infected cells or latent viral antigens presented by antigen-presenting cells. This approach was hypothesized to enhance the persistence and expansion of the CAR-T cells in vivo, addressing a major limitation of first-generation CAR constructs in pediatric solid tumors like neuroblastoma and osteosarcoma.
