Overview

157cysAP1-PBNC is a designed protein-cage nanoparticle that displays ligand-specific peptide bunches (PBNCs) for high affinity and biological activity. It is an AP-1-containing nanocage protein (AP1-PBNC) formed by genetically fusing the interleukin-4 receptor (IL-4R)-targeting peptide, AP-1, with ferritin-L-chain (FTL). The FTL undergoes 24-subunit assembly to create a highly stable nanocage. This design allows for the clustering of AP-1 peptides on the nanocage surface, synergistically increasing their avidity for IL-4R. 157cysAP1-PBNC specifically binds to IL-4R-expressing cells and has demonstrated enhanced binding avidity compared to the free AP-1 peptide. In a murine model of experimental asthma, treatment with AP1-PBNCs diminished airway hyper-responsiveness, eosinophilic airway inflammation, and decreased mucus hyperproduction, indicating its potential as a therapeutic agent for diseases like asthma and cancer.
