**15cl12-MMAE** is a preclinical, site-specifically conjugated anti-immunoglobulin E antibody-drug conjugate developed at the University of Natural Resources and Life Sciences Vienna. It comprises the chimeric anti-membrane immunoglobulin E antibody 15cl12, engineered with an L328C cysteine for conjugation of monomethyl auristatin E through a maleimidocaproyl-valine-citrulline-p-aminobenzoyloxycarbonyl linker. The antibody recognizes the extracellular membrane-proximal domain of membrane-bound immunoglobulin E on immunoglobulin E-positive cells, undergoes rapid target-cell internalization, and delivers the tubulin-inhibitory payload to induce cytotoxicity. The construct had a drug-to-antibody ratio of 2 and showed cytotoxic activity against an immunoglobulin E-positive cell line, supporting potential development as a cell-depleting therapy for immunoglobulin E myeloma and immunoglobulin E-associated diseases.
