Overview

161Tb-LNC1011 is a prostate-specific membrane antigen (PSMA)-targeted radioligand therapy developed by Peking Union Medical College Hospital for the treatment of metastatic castration-resistant prostate cancer (mCRPC). The drug consists of the LNC1011 peptide, which features a PSMA-targeting moiety, a DOTA chelator, and a dansyl moiety designed for albumin binding to prolong blood circulation and enhance tumor uptake. It is radiolabeled with terbium-161 ($^{161}$Tb), a radioisotope that emits beta particles similar to lutetium-177 but also a high proportion of low-energy Auger and conversion electrons. These Auger electrons have an ultra-short tissue range (< 500 nm) and high linear energy transfer, potentially providing superior efficacy in treating small clusters of cancer cells or micrometastases compared to pure beta-emitters. The drug is currently being evaluated in Phase 1 clinical trials for safety, dosimetry, and preliminary efficacy.
