177Lu-4A06 is a CDCP1-targeted radiopharmaceutical consisting of the monoclonal antibody 4A06 labeled with the beta-emitting radioisotope Lutetium-177. It is designed to treat cancers that overexpress CUB domain-containing protein 1 (CDCP1), such as the basal/squamous subtype of bladder cancer. The 4A06 antibody binds to the extracellular domain of CDCP1, allowing for the targeted delivery of ionizing radiation to tumor cells, which results in DNA damage and subsequent cell death. Preclinical studies have shown that 177Lu-4A06 effectively inhibits tumor growth in bladder cancer xenograft models, including those that do not express other common targets like TROP2 or NECTIN4.
