177Lu-9E7.4 is a radioimmunotherapy (RIT) agent currently under investigation for the treatment of multiple myeloma. It consists of the murine monoclonal antibody 9E7.4, which specifically targets the CD138 (syndecan-1) antigen, a protein highly expressed on the surface of malignant plasma cells. The antibody is conjugated to Lutetium-177 (177Lu), a radioisotope that emits beta particles with a maximum energy of 0.5 MeV and a half-life of 6.7 days. This allows for the delivery of targeted cytotoxic radiation directly to tumor cells, causing double-stranded DNA breaks and subsequent apoptosis. Developed primarily by research teams in Nantes, France (including INSERM and the University of Nantes), 177Lu-9E7.4 has been evaluated in preclinical models to compare its therapeutic efficacy against alpha-emitting isotopes like Bismuth-213, aiming to optimize radioimmunotherapy strategies for hematologic malignancies.
