Drug intelligence / Profile preview
177Lu-ABY-027 (also known as [177Lu]Lu-ABY-027) is a preclinical-stage therapeutic radiopharmaceutical developed by Affibody AB for the treatment of HER2-expressing cancers, such as breast and ovarian cancers. The molecule is a fusion protein consisting of a second-generation HER2-targeting Affibody molecule (ZHER2:2891) fused to a high-affinity albumin-binding domain (ABD035). It is site-specifically conjugated at the C-terminus with a DOTA chelator and labeled with the beta-emitting radionuclide lutetium-177 (177Lu). The inclusion of the ABD extends the circulatory half-life of the molecule by binding to serum albumin, which prevents glomerular filtration and significantly reduces renal uptake, thereby optimizing the biodistribution profile for targeted radionuclide therapy. Preclinical studies have demonstrated that 177Lu-ABY-027 effectively targets HER2-expressing tumors and extends survival in animal models, both as a monotherapy and in combination with trastuzumab.
