Drug intelligence / Profile preview
177Lu-B7-H3 ADC is a preclinical radiolabeled antibody-drug conjugate (ADC) developed by Peking University First Hospital for the treatment of B7-H3-expressing tumors, particularly prostate cancer. The agent consists of a human monoclonal antibody targeting B7-H3 (CD276) conjugated to a topoisomerase inhibitor payload, which is further radiolabeled with the beta-emitting radionuclide lutetium-177 (177Lu). By combining the targeted cytotoxic mechanism of the ADC with the localized radiation of radioligand therapy, 177Lu-B7-H3 ADC achieves synergistic antitumor efficacy. Preclinical studies in prostate cancer xenograft models have demonstrated significant tumor growth inhibition and survival benefits with manageable and reversible hematological toxicity.
