177Lu-daratumumab is a radiopharmaceutical agent composed of the human monoclonal antibody daratumumab labeled with the beta-emitting radioisotope lutetium-177 (177Lu). Daratumumab specifically targets CD38, a transmembrane glycoprotein overexpressed in multiple myeloma and various types of lymphoma. By binding to CD38-positive cells, the compound delivers localized ionizing radiation from the 177Lu isotope, which has a half-life of approximately 6.7 days and a tissue penetration range of about 2 mm. This targeted approach, known as radioimmunotherapy (RIT), aims to induce tumor cell death while minimizing damage to surrounding healthy tissue. The gamma emissions of 177Lu also allow for theranostic applications, enabling non-invasive imaging and dosimetry. It is currently being investigated in preclinical models for hematological malignancies.
