177Lu-DOTA-30F11 is an experimental radiopharmaceutical designed for the treatment of hematologic malignancies, specifically acute myeloid leukemia (AML). It consists of the rat IgG2b monoclonal antibody 30F11, which targets the CD45 antigen (leukocyte common antigen) expressed on the surface of most hematopoietic cells and leukemic blasts. The antibody is conjugated to the beta-emitting radionuclide Lutetium-177 (177Lu) using the DOTA (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid) chelator. While 177Lu offers favorable imaging properties due to its low-energy gamma emissions and a shorter path length compared to Yttrium-90, preclinical studies in murine models have shown that 177Lu-DOTA-30F11 may be less effective than its 90Y counterpart in treating disseminated leukemia, potentially due to differences in absorbed radiation doses and dose rates.
