Drug intelligence / Profile preview
177Lu-DOTA-HYNIC-panPSMA (also known as TLX597-Tx or TLX597) is an investigational, next-generation small-molecule radioligand therapy (RLT) being developed by Telix Pharmaceuticals. The drug consists of the beta-emitting radionuclide lutetium-177 (177Lu) chelated by DOTA and conjugated to a HYNIC-linked panPSMA ligand. It is designed to target prostate-specific membrane antigen (PSMA), which is highly overexpressed on prostate cancer cells. Upon binding, the radiopharmaceutical is internalized, delivering targeted ionizing radiation that induces DNA damage and cell death. Due to its optimized design, 177Lu-DOTA-HYNIC-panPSMA exhibits a highly favorable dosimetry profile with significantly reduced uptake in healthy organs, such as the salivary glands and kidneys, potentially minimizing off-target toxicities like xerostomia and renal impairment. It is currently being evaluated in Phase 2 clinical trials, including the OPTIMAL-PSMA study for metastatic castration-resistant prostate cancer (mCRPC) and the OPTIMAL-e study for metastatic hormone-sensitive prostate cancer (mHSPC).
