177Lu-DOTA-rat IgG is a radiolabeled isotype control antibody used in preclinical radioimmunotherapy research. It consists of polyclonal rat immunoglobulin G (IgG) conjugated to the chelator DOTA (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid) and labeled with the beta-emitting radionuclide Lutetium-177. In experimental models of hematologic malignancies, such as acute myeloid leukemia, it serves as a non-targeting negative control to differentiate the therapeutic effects and toxicities of antigen-specific radiolabeled antibodies (e.g., anti-CD45) from non-specific radiation exposure. Because it does not bind to specific cell surface antigens in the murine models described, its biodistribution reflects blood pool clearance and non-specific uptake, providing a baseline for evaluating the efficacy of targeted radioimmunotherapy.
