177Lu-DOTA-SNA040 is a targeted radiopharmaceutical therapeutic candidate developed by Suzhou SmartNuclide Biopharmaceutical. It consists of a ligand (likely a single-domain antibody or nanobody) that specifically binds to **Claudin 18.2 (CLDN18.2)**, conjugated to the beta-emitting radioisotope **Lutetium-177 (177Lu)** via a DOTA chelator. Claudin 18.2 is a tight junction protein that is highly overexpressed in several gastrointestinal cancers, including gastric, pancreatic, and esophageal tumors, while remaining largely absent in healthy tissues. Upon binding to the target cells, the drug delivers localized ionizing radiation, leading to DNA damage and cell death in the tumor mass. It is currently being investigated in first-in-human Phase 1 clinical trials for patients with advanced CLDN18.2-positive solid tumors who have failed standard therapies.
