177Lu-DOTA-tTF is a radiolabeled fusion protein drug candidate consisting of tissue factor (tTF) site-specifically conjugated to the chelator DOTA and labeled with the therapeutic radioisotope lutetium-177 (177Lu). It is designed for targeted tumor therapy: the tTF entity binds to and induces thrombosis in tumor vasculature, thereby blocking blood supply to the tumor, while 177Lu delivers localized beta radiation to maximize tumor cell kill. This dual-action approach is considered a novel antitumor strategy, combining vascular targeting with radiotherapeutic effects. Preclinical and early clinical studies have focused on solid tumors expressing an accessible tumor vasculature.
