177Lu-DOTA-zolbetuximab is a claudin 18.2-targeted radiopharmaceutical therapy designed for the treatment of gastric and pancreatic cancers. It consists of the monoclonal antibody zolbetuximab, which specifically binds to claudin 18.2 (CLDN18.2), conjugated via a DOTA chelator to the beta-emitting radioisotope lutetium-177 (177Lu). This agent delivers targeted radiation directly to tumor cells expressing CLDN18.2, a biomarker frequently overexpressed in upper gastrointestinal malignancies. Preclinical studies have demonstrated significant tumor regression and complete eradication in some models without observed radiation-induced toxicity.
