177Lu-DOTAGA-IAB56 is a radiopharmaceutical therapy (RPT) agent consisting of a minibody (IAB56) conjugated to the chelator DOTAGA and radiolabeled with the beta-emitting isotope Lutetium-177 (177Lu). The minibody is an 80 kDa antibody fragment composed of two scFv antigen-binding domains fused to the CH3 domain of human IgG, designed for rapid blood clearance and high target affinity. It specifically targets integrin alpha-V beta-6 (αVβ6), a cell surface protein that is significantly upregulated in various carcinomas, including pancreatic ductal adenocarcinoma, while remaining largely absent in healthy adult tissues. Developed by ImaginAb, this agent aims to deliver targeted radiation to tumor cells while minimizing hematological and off-target organ exposure. Preclinical studies in pancreatic cancer xenograft models have demonstrated significant tumor growth inhibition and improved survival.
