177Lu-FC706 is a therapeutic radionuclide drug conjugate (RDC) developed by FindCure Biosciences. It consists of a targeting ligand directed against Nectin cell adhesion molecule 4 (Nectin-4) conjugated to the beta-emitting radioisotope Lutetium-177 (177Lu). Nectin-4 is a cell surface adhesion molecule highly overexpressed in various solid tumors, including urothelial, breast, lung, and pancreatic cancers, while showing limited expression in normal tissues. Upon binding to Nectin-4 on tumor cells, 177Lu-FC706 delivers localized, high-energy beta radiation, inducing DNA double-strand breaks and subsequent cell death in both targeted cells and neighboring tumor cells via the bystander effect. The drug is currently in preclinical and IND-enabling development for the treatment of Nectin-4-positive advanced solid tumors, with a companion diagnostic imaging agent, 68Ga-FC706, being developed in parallel for patient selection and dosimetry.
