Drug intelligence / Profile preview
177Lu-G3-ABD is a preclinical radiopharmaceutical designed for targeted radionuclide therapy of human epidermal growth factor receptor 2 (HER2)-expressing malignancies. The agent consists of the HER2-targeting designed ankyrin repeat protein (DARPin) G3 fused at its C-terminus to an albumin-binding domain (ABD), conjugated with a DOTA chelator, and radiolabeled with the beta-emitting isotope lutetium-177 (177Lu). While small scaffold proteins like DARPins typically suffer from rapid renal clearance and high kidney reabsorption, the fusion of the ABD allows the construct to bind to serum albumin in circulation. This interaction significantly extends the blood half-life, reduces renal accumulation, and enhances tumor uptake, thereby optimizing the therapeutic index for treating HER2-positive cancers such as breast and ovarian cancers.
