177Lu-GT-008 is a preclinical-stage targeted radiotherapeutic candidate consisting of the glycan-dependent humanized IgG1 monoclonal antibody GT-008 conjugated and radiolabeled with the beta-emitting radioisotope lutetium-177 (177Lu). Developed by Pentixapharm (originally discovered via Glycotope's GlycoTarget platform), GT-008 selectively targets a tumor-associated O-glycoform of CD24 (specifically carrying the Thomsen-Friedenreich [TF] core-1 antigen) that is highly prevalent on solid tumors but has limited expression on healthy tissues. This glycoform-selective binding allows 177Lu-GT-008 to deliver cytotoxic beta radiation directly to cancer cells while sparing healthy CD24-expressing immune cells and tissues. Preclinical studies in breast cancer xenograft models have demonstrated robust tumor growth control, favorable biodistribution, and complete remissions.
