Overview

IM-3050 is a fibroblast activation protein (FAP)-targeted radioligand therapy (RLT) being developed by Immunome, Inc. for the treatment of advanced solid tumors. The therapeutic candidate consists of a small molecule FAP-specific ligand conjugated to the beta-emitting radioisotope lutetium-177 (177Lu). Its molecular architecture includes four functional domains: the FAP-targeting ligand, a linker tuned for tumor-specific uptake, an albumin-binding domain designed to enhance tumor retention and improve the therapeutic index, and a chelator for the radionuclide. FAP is a cell surface protease highly expressed on cancer-associated fibroblasts (CAFs), which are prevalent in the stroma of approximately 75% of solid tumors. By targeting the tumor stroma, IM-3050 aims to deliver localized radiation to the tumor microenvironment. Immunome submitted an Investigational New Drug (IND) application to the FDA in March 2025, with Phase 1 clinical trials expected to commence in the second half of 2025.
