Drug intelligence / Profile preview
**177Lu-NNV003** (Humalutin) is a radiopharmaceutical radioimmunoconjugate designed for the treatment of B-cell malignancies, most notably non-Hodgkin’s lymphoma (NHL). It consists of a chimeric mouse-human monoclonal antibody (NNV003) that targets **CD37**, a surface antigen highly expressed on B cells, conjugated with the beta-emitting radionuclide **lutetium-177** (using p-SCN-Bn-DOTA as the chelator). Upon binding to CD37-positive malignant B cells, the antibody delivers targeted radiation that induces cytotoxicity in tumor tissue while sparing much of the surrounding normal tissue. The antibody component also engages immunological mechanisms such as antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP). Preclinical models and early animal studies have demonstrated significant anti-tumor efficacy and favorable toxicity profiles, with reversible hematological toxicity as the main side effect. 177Lu-NNV003 is being evaluated for use in various B-cell malignancies, including **diffuse large B-cell lymphoma (DLBCL)**, **mantle cell lymphoma (MCL)**, and **chronic lymphocytic leukemia (CLL)**. Its different antigenic target (CD37) compared to the more commonly targeted CD20 may provide an option for patients with disease refractory to anti-CD20 therapies[1][2][3][4][6].
