Overview

177Lu-PNT2002 + pembrolizumab is an investigational combination therapy being evaluated for the treatment of metastatic clear cell renal cell carcinoma (ccRCC). 177Lu-PNT2002, also known as 177Lu-PSMA I&T, is a radiopharmaceutical that targets the prostate-specific membrane antigen (PSMA), which is expressed on the neovasculature of various solid tumors, including RCC. It delivers the beta-emitting radioisotope lutetium-177 directly to the tumor site. Pembrolizumab is a well-established immune checkpoint inhibitor that targets the programmed cell death protein 1 (PD-1) receptor. The combination is being studied by the Sidney Kimmel Comprehensive Cancer Center at Johns Hopkins in a Phase 1b/2 trial (NCT05973110) to determine if the localized radiation from the radioligand therapy can enhance the efficacy of immunotherapy in patients who have previously progressed on anti-PD-1 or PD-L1 treatments.
