17G8 is an afucosylated monoclonal antibody developed by AbbVie that targets Immunoglobulin-like transcript 3 (ILT3), also known as LILRB4. ILT3 is an inhibitory receptor expressed on mature monocytes, dendritic cells (DC), plasma blasts, and various hematopoietic malignant cells, including those in acute myeloid leukemia (AML), B cell lymphoma, and multiple myeloma (MM). By blocking ILT3, 17G8 promotes the maturation of dendritic cells and stimulates the production of interferon-gamma (IFNγ), which in turn enhances T cell-mediated anti-tumor immunity. The antibody is engineered with enhanced Fc function through afucosylation to further reduce tumor burden independently of traditional antibody-dependent cellular cytotoxicity (ADCC). Preclinical studies have demonstrated that 17G8 can impair the dissemination of AML cells and, when combined with PD-1 inhibitors, significantly increase CD8+ effector memory T cells and improve survival outcomes.
