Drug intelligence / Profile preview
The 18-component perfusion solution is an experimental extracellular-type organ preservation solution designed specifically for hypothermic oxygenated machine perfusion (HOPE) of donor organs, such as the liver and kidney, during transplantation. Developed by researchers at the NV Sklifosovsky Research Institute of Emergency Medicine, the solution is formulated on an extracellular electrolyte platform (containing 100 mM sodium and 5 mM potassium) and incorporates active components targeting five key ischemia-reperfusion injury (IRI) pathways. Specifically, it contains a competitive mitochondrial reverse electron transport (RET) inhibitor, a mitochondrial permeability transition pore (mPTP) stabilizer (FKBP12-mediated), an antioxidant module (glutathione precursor and amphiphilic antioxidant), an antiferroptotic module (high-affinity Fe3+ chelator and GPX4 cofactor), an immunomodulator, and energy substrates. In preclinical models of donation after circulatory death (DCD), the solution has demonstrated significant hepatoprotective and nephroprotective effects, reducing hepatocellular injury, suppressing ferroptosis, and improving bile production and mitochondrial viability.
