Overview

18A3 is a mouse monoclonal antibody targeting Serum amyloid A1 (SAA1), developed by researchers at Columbia University. SAA1 is a pro-inflammatory protein that is significantly elevated in the bone marrow of patients with myelodysplastic syndromes (MDS) and acute myeloid leukemia (AML), where it promotes the expansion of leukemic stem cells and malignant clones through the activation of NFκB signaling and the induction of pro-inflammatory cytokines (e.g., IL-6, TNF-α). 18A3 acts by neutralizing SAA1, thereby inhibiting the proliferation of malignant cells and disrupting the inflammatory niche that supports disease progression. Preclinical studies have demonstrated its efficacy in reducing leukemic burden and improving survival in AML mouse models, as well as inhibiting growth in solid tumor lines such as pancreatic cancer.
