Drug intelligence / Profile preview
18F-FPBT is a novel fluorine-18-labeled positron emission tomography (PET) radiotracer designed for the non-invasive, direct detection of oxidative stress in vivo. Chemically, it is [18F]3-(3-fluoropropyl)-2-phenyl-2,3-dihydrobenzo[d]thiazole. The tracer is highly selective for oxidation by superoxide compared to other physiological reactive oxygen species (ROS). In its reduced form, 18F-FPBT is lipophilic, allowing it to penetrate cell membranes and the blood-brain barrier. Upon reaction with intracellular superoxide, it is oxidized to a hydrophilic form that is transiently retained within cells, providing ROS-sensitive PET contrast. Developed by King's College London and the First Affiliated Hospital of Dalian Medical University, 18F-FPBT has demonstrated preclinical efficacy in visualizing cardiac oxidative stress in animal models of doxorubicin-induced cardiotoxicity and lipopolysaccharide (LPS)-induced inflammation, and is undergoing validation for clinical translation.
