Drug intelligence / Profile preview
18F-OncoFAP is a fluorine-18 labeled small molecule radiopharmaceutical designed for positron emission tomography (PET) imaging of various solid tumors. It targets fibroblast activation protein (FAP), a type II transmembrane glycoprotein that is highly overexpressed by cancer-associated fibroblasts (CAFs) in the tumor stroma of most epithelial cancers, while remaining nearly undetectable in healthy adult tissues. The ligand, OncoFAP, is an ultra-high-affinity organic molecule with a dissociation constant (Kd) of approximately 680 pM. The 18F-labeled version is typically synthesized using an aluminum fluoride ([18F]AlF) complexing strategy with a NOTA chelator, allowing for efficient production and high radiochemical yields. Developed through a partnership between Philochem and Bracco (via Blue Earth Diagnostics), 18F-OncoFAP serves as a pan-cancer diagnostic tool to visualize the tumor microenvironment and potentially guide FAP-targeted therapies.
