Overview

18F-THK5317 is a radiopharmaceutical used as a positron emission tomography (PET) imaging agent for the in vivo detection of tau protein aggregates in the brain. It is primarily developed as a biomarker for Alzheimer's disease and related tauopathies. The compound binds to neurofibrillary tangles composed of hyperphosphorylated tau protein, allowing visualization and quantification of tau pathology in patients. Studies have shown that 18F-THK5317 can discriminate between healthy controls, prodromal Alzheimer's disease, and Alzheimer's dementia groups with high accuracy. Additionally, early-phase imaging with this tracer provides information on brain perfusion closely related to glucose metabolism. The tracer has been evaluated head-to-head against other tau PET tracers such as 18F-flortaucipir and has demonstrated strong predictive value for cognitive decline in Alzheimer’s disease patients[1][5][6][7].
