18F-uPA-probe is a fluorine-18 labeled activity-based positron emission tomography (PET) imaging probe designed for the non-invasive visualization of active urokinase plasminogen activator (uPA) expression in tumors. Developed by researchers at the University of Antwerp, the probe is based on the small-molecule, irreversible, and selective uPA inhibitor UAMC-00150. It consists of a diphenyl phosphonate warhead with a guanidine phenylalanine moiety at the P1 position, connected via a polyethylene glycol (PEG) linker to a [18F]-4-fluorobenzoyl group. The probe covalently and irreversibly binds to the active site of uPA, allowing quantitative PET imaging of uPA activity, which is a key biomarker for cancer invasion and metastasis (particularly in breast cancer).
