Overview

19(T2)28z-1XX CAR T cells are an autologous **chimeric antigen receptor (CAR) T cell therapy** specifically engineered to target **CD19**, a protein found on the surface of B-cell malignancies. This therapy features an innovative “1XX” signaling domain, which is an optimized mutant of the CD3ζ chain containing just one immunoreceptor tyrosine-based activation motif (iTAM) rather than the conventional three. The design, which also includes CD28 costimulatory signaling, is intended to **modulate and calibrate CAR T cell activation**, enabling potent anti-tumor effects at **lower cell doses** while reducing the risk and severity of cytokine release syndrome (CRS) and immune effector cell-associated neurotoxicity syndrome (ICANS). Clinical trial data in **relapsed/refractory large B-cell lymphoma** indicate high overall response and complete response rates with a favorable safety profile and potentially durable remissions. The platform may have broader applications in other hematologic malignancies, solid tumors, and autoimmune diseases[1][3][5][7][9].
