19-22 is an investigational bispecific chimeric antigen receptor (CAR) T-cell therapy being developed by Cellogen Therapeutics for the treatment of B-cell malignancies, specifically Acute Lymphoblastic Leukemia (ALL) and Diffuse Large B-Cell Lymphoma (DLBCL). The therapy is engineered to simultaneously target two distinct antigens, CD19 and CD22, to overcome the challenge of antigen-escape relapse, which occurs when cancer cells lose the CD19 target. Additionally, the 19-22 construct incorporates dual costimulatory domains (utilizing combinations of CD28, 4-1BB, ICOS, and OX40) designed to enhance the persistence and anti-tumor activity of the CAR T cells. Currently in preclinical development, the program aims to address the limitations of first- and second-generation CAR T therapies regarding long-term efficacy and durability of response.
