19-28z-1A is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target CD19-expressing malignancies. Developed by researchers at Columbia University and Memorial Sloan Kettering Cancer Center, this construct incorporates a unique pTα 1A domain derived from the pre-T cell receptor alpha chain, alongside a CD28 costimulatory domain and a CD3ζ signaling domain. The inclusion of the pTα 1A domain is intended to enhance CAR T-cell sensitivity against tumors with low antigen density, improve T-cell expansion and cytokine production, and increase in vivo persistence while mitigating exhaustion. Preclinical studies have demonstrated that 19-28z-1A can achieve long-term tumor clearance in CD19-low models (such as NALM6 variants) where standard 28z or 1XX CAR architectures typically fail.
