19-28z CAR EBV-VSTs is an allogeneic, "off-the-shelf" chimeric antigen receptor (CAR) T-cell therapy developed by Memorial Sloan Kettering Cancer Center. The product utilizes Epstein-Barr virus (EBV)-specific T cells (EBV-VSTs) as a vehicle, which are genetically modified via gammaretroviral transduction to express a second-generation CAR targeting the CD19 antigen. This specific CAR construct, known as 19-28z, incorporates a CD28 costimulatory domain and a CD3-zeta signaling domain to enhance T-cell activation, proliferation, and persistence. By using EBV-VSTs derived from healthy donors, the therapy aims to reduce the risk of graft-versus-host disease (GVHD) and allows for the creation of a pre-manufactured cell bank, providing a readily available treatment for patients with relapsed or refractory B-cell malignancies without the delays associated with autologous manufacturing.
